Tspan3 was identified as a novel ADAM10 interaction partner. Tspan3 modulates ectodomain shedding and intramembrane proteolysis of ADAM10. Tspan3 increases ADAM10-mediated generation of APP α-CTFs and reduces Aβ liberation. Tspan3 stabilizes ADAM10, APP and the γ-secretase complex at the plasma membrane Deficiency of Tspan3 is (most likely) compensated by other tetraspanins.
