Hemoglobin (Hb) was successfully combined with carboxyl graphene (CG) through condensation reaction to form novel bio-nanocomposite hemoglobin-carboxyl graphene bio-nanocomposites (CG-Hb). Scanning electron microscope (SEM), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), Fourier transform infrared spectroscopy (FTIR) and Raman spectra indicated that the morphology and structure of CG-Hb. Cyclic voltammograms of CG-Hb modified on the gold nanoparticles (Au-NPs) electro-deposited on glassy carbon electrode (GCE) exhibited a pair of redox peaks (−0.235 V and −0.122 V vs. SCE) and showed excellent biosensor for hydrogen peroxide, which realized the direct electron transfer between CG-Hb and GCE. The electrochemistry properties and the ability of oxygen-carrying were investigated by electrochemical methods. It was found this kind of bio-nanocomposite maintained the natural biological activity of Hb and was explored to be no observable denaturation, which can be considered as a novel effective oxygen carrier in electrochemical aspect for further research.
