Cell refractive index mapping is realized in a focused scanning structure based on the polarization-sensitive light absorption of graphene. Large longitudinal detection depth of the graphene evanescent field enables high quality cell refractive index measurement. Perfect optical vortex beam is theoretically simulated and experimentally generated, to control the focusing angles. Refractive index difference of two human colonic cancer cells is obtained from mapping image by the proposed method, which could be applied in the early diagnosis and prevention of cancer.
