Sucrose isomerase and oligo-1,6-glucosidase (O16G) belongs to the GH13_31 subfamily. Azotobacterglucocosidase (AZOG) is intermediary SIase harboring both RLDRD motif and O16G-like conserved region. AZOG exhibited O16G similar substrate specificity and intermolecular transglycosylation ability to synthesize isomaltose using methyl-α-glucoside. Shuffling with SIase resulted in change of substrate specificity of AZOG and revival of sucrose isomerase activity.
