Free-floating 3D cultures from hESC acquire a high degree of differentiation. Neurons, astrocytes and oligodendrocytes are organized in a histotypic manner. Electrophysiological measurements showed different firing patterns. Acute treatment with known neurotoxicants upregulated HMOX and GFAP expression. These 3D cultures could be a useful model for developmental neurotoxicity testing.
