Small-volume diagnostic blood samples are an underexploited source of HSPCs. ACK lysis and density-gradient separation are equally efficient for WBC isolation. Gradient isolation and 50% FBS are recommended for biorepository creation. HSPCs from fresh and frozen samples are expandable in vitro without enrichment. Isolated HSPCs are suitable for lentiviral transduction and differentiation.
