The PYC gene from Penicillium viticola 152 was cloned and characterized by Genome Walking method. An ORF of 3582 bp encoding 1193 amino acids protein (isoelectric point: 5.01) was characterized. A promoter of gene contained a TATA box, CAAT boxes, the 5′-SYGGRG-3′ and a 5′-HGATAR-3′ sequences. The protein deduced from the PYC gene was a homotetramer (4), and had four functional domains. The specific pyruvate carboxylase activity was much higher in medium with CSL than that of biotin.
