Tyrosine kinase receptor c-ros-oncogene 1 (C-ROS-1) is identified as a novel downstream Twist-1 target in human BMSC. Twist-1 negatively regulates expression of C-ROS-1 in BMSC. Knock-down of c-ros-1 results in reduced osteogenic potential, increased adipogenic potential and an increase in PI3K/AKT/mTORC1 signalling. c-ros-1 and PI3K/AKT/mTORC1 signalling are novel, Twist-1 downstream mechanisms, involved in BMSC fate switching between osteogenesis and adipogenesis.
