A mutation system for Tetrahymena outer arm dynein has been established. The P-loop mutations are stably maintained in DYH3-knockout cells. The P3 mutation does not affect Dyh3p localization but abolishes Dyh3p motility. No significant effect of the P4 mutation on Dyh3p function was detected. No P1 or P2 mutant cells were isolated from DYH3-knockout cells.
