A shell particle consists of a solid, nonporous core thatis surrounded with a shell of a porous solid havingessentially the same physicochemical properties as thoseof the conventional porous particles used as packingmedia in chromatography. The diameter of the solid coreand the thickness of its shell or the external diameter ofthe particle characterizes the chromatographic propertiesof the packing material. The potential advantage of thisparticle structure would be the shorter average path lengthexperienced by solute molecules during their diffusionacross the particles of packing material when they areretained. Compounds having slow pore diffusion wouldexhibit higher efficiencies on columns packed with shellthan with conventional, fully porous particles. Usingcolumns packed with Halo, a new type of porous silicashell particles, we assess the gain achieved with thisprinciple for peptides of moderate molecular weights andfor small proteins.
