The molecu1ar beacon (MB) is a new fluorescence probecontaining a single-stranded oligonucleotide with a probesequence embedded in complementary sequences thatform a hairpin stem. Due to the inherent fluorescent signaltransduction mechanism, an MB functions as a sensitiveprobe with a high signal-to-background ratio for real-timemonitoring and provides a variety of exciting opportunitiesin DNA, RNA, and protein studies. To better understandthe properties of MBs, the specific interactions betweenMB and target DNA (complementary and one-base mismatch) have been directly investigated by atomic forcemicroscopy. The interaction force between a linear DNAprobe and the target DNA was also detected and compared to that between MB and target DNA. The resultsdemonstrate the high specificity of the MB/target DNAcompared to the linear DNA/target DNA interaction.
