中药诃子抑制金黄色葡萄球菌生物被膜形成机制研究

摘要
目的探讨中药诃子对金黄色葡萄球菌(金葡菌)生物被膜形成的抑制作用,为临床联合用药提供理论依据和指导。方法首先检测诃子水提物对游离态金葡菌的抑制效果,之后在建立生物被膜诱导模型基础上评估诃子对生物被膜态金葡菌的抑制效果;进一步评估诃子水提物对金葡菌生物被膜的干预作用,在此基础上探索诃子联合红霉素对金葡菌的抑制效果。结果体外抑菌实验结果显示,诃子水提物对浮游态金葡菌的最小抑菌浓度和最小杀菌浓度分别为3.90mg/ml和31.20mg/ml,对生物被膜态金葡菌的最小抑菌浓度和最小杀菌浓度分别为15.60mg/ml和62.50mg/ml,均具有较好的杀伤效果。生物被膜测定结果显示,诃子水提物对金葡菌生物被膜形成具有抑制作用,呈剂量依赖性。体外联合用药的结果表明,联合使用一定浓度的诃子水提物、红霉素对金葡菌的抑制和杀伤作用显著增强。结论中药诃子对金葡菌生物被膜形成有抑制作用,与红霉素联用能够增强其对金葡菌的杀伤作用,本研究为临床上金葡菌感染的诊治提供了理论指导。
        OBJECTIVE To explore the effect of Chinese herb myrobalan on inhibiting the formation of biofilms in Staphylococcus aureus so as to provide guidance for combined use of antibiotics.METHODS Firstly,the effect of herb myrobalan aqueous extracts on S.aureus in suspension was tested,and the inhibitory effect on S.aureus in biofilm state was also examined after the biofilm-induced model was established,the intervention effect of the herb myrobalan aqueous extracts on the S.aureus biofilm was further evaluated,and the inhibitory effect of herb myrobalan combined with erythromycin was explored.RESULTS The result of in vitro bacteriostatic test showed that the minimal inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of herb myrobalan aqueous extracts on S.aureus in suspension were 3.90 mg/ml and 31.20 mg/ml,respectively,the MIC and MBC on S.aureus in biofilm state were 15.60 mg/ml and 62.50 mg/ml,respectively.The result of biofilm test indicated that the herb myrobalan aqueous extracts had inhibitory effect on formation of the S.aureus biofilms in a dose-dependent way.The result of in vitro combined medication showed that erythromycin combined with herb myrobalan aqueous extracts had remarkable inhibitory and lethal effect on the S.aureus.CONCLUSION The herb myrobalan has the inhibitory effect on formation of S.aureus biofilms,and its combination with erythromycin may intensify the lethal effect on the S.aureus strains,which provide theoretical basis for clinical diagnosis and treatment of the S.aureus infection.
