猴耳环正丁醇部位抗类风湿性关节炎的作用及机制研究

摘要
目的探讨猴耳环正丁醇部位抗类风湿性关节炎的作用及机制。方法采用猴耳环水提总浸膏制备其正丁醇部位。将48只SD大鼠随机分为6组,分别为空白组,模型组,阳性药组(雷公藤多苷片,0.01 g·kg~(-1)),猴耳环高(H)、中(M)、低(L)剂量组(14、7、3.5 g·kg~(-1));采用注射弗氏完全佐剂诱发大鼠佐剂性关节炎模型;每日1次,连续灌胃给药28 d。记录大鼠体质量变化;测定大鼠原发侧、继发侧足肿胀容积变化;测定并计算大鼠脾脏指数;检测全血及血浆黏度;ELISA法测定大鼠血清中白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、前列腺素E_2(PGE_2)水平;HE染色观察大鼠踝关节病理组织形态学变化。结果与空白组比较,模型组大鼠原发侧及继发侧足肿胀度,脾脏指数,全血(低切)及血浆黏度,大鼠血清中炎症因子IL-1β、TNF-α、PGE_2水平均明显升高(P <0.05,P <0.01)。与模型组比较,猴耳环H、M组的原发侧足肿胀度、脾脏指数明显降低(P<0.01,P<0.05);猴耳环H组的全血(低切)及血浆黏度、血清TNF-α水平明显降低(P<0.05,P<0.01);猴耳环各剂量组的继发侧足肿胀度、血清IL-1β及PGE2水平明显降低(P<0.05,P<0.01),关节组织的炎细胞浸润减少、病理形态得到改善。结论猴耳环正丁醇部位具有抗类风湿性关节炎作用,其作用机制可能与降低血清炎症因子IL-1β、TNF-α、PGE_2的含量及改善血液流变学有关。
        Objective To screen the effective parts for anti-rheumatoid arthritis from Pithecellobium clypearia and to explore its mechanism. Methods The n-butanol fraction was prepared from crude aqueous extract of Pithecellobium clypearia. The rheumatoid arthritis model rats were randomly divided into 6 groups: blank group, model group,positive group(Tripterygium wilfordii tablets),Pithecellobium clypearia n-butanol fraction high(H),medium(M)and low(L)dose groups(14 g·kg~(-1),7 g·kg~(-1),and 3.5 g·kg~(-1),respectively). Rats were observed for 28 days. Body weights of the rats were recorded,and changes of swelling degrees of left and right hind feet were recorded. After the treatment, the spleen indexes of the rats were calculated. The changes of whole blood and plasma viscosity,histopathology,and serum contents of interleukin(IL)-1β,tumor necrosis factor-α(TNF-α),prostaglandia E_2(PGE_2)were measured. Results Compared with the blank control group,the levels of primary and secondary foots welling,spleen index,whole blood(low-shear)and plasma viscosity,and inflammatory factors IL-1β,TNF-α and PGE_2 in the serum of the model group rats were significantly increased(P < 0.05,P < 0.01). Compared with the model group,the degree of primary foot swelling and spleen indexes were significantly reduced in the positive drug group,Pithecellobium clypearia H and M groups(P < 0.01,P < 0.05). The whole blood(low-shear)and plasma viscosity, serum TNF-α content of the Pithecellobium clypearia H group were significantly decreased(P < 0.05,P < 0.01). The degree of secondary lateral foot swelling,serum levels of inflammatory cytokines IL-1β and PGE_2 were significantly reduced in Pithecellobium clypearia H,M,and L groups(P < 0.05,P < 0.01);the infiltration of inflammatory cells in the joint tissue was reduced and the morphology was improved. Conclusion The n-butanol fraction of Pithecellobium clypearia has anti-rheumatoid arthritis effect. Its mechanism may be through reducing the contents of IL-1β,TNF-α and PGE_2 in serum and improving the hemorrheology.
