摘要
目的:改进《中国药典》色谱条件,建立HPLC法测定复方甘草片中甘草酸含量测定的方法。方法:Shim-pack GIST C_(18)色谱柱(4.6 mm×250 mm,5μm),流动相:0.025 mol·L~(-1)磷酸二氢钾溶液-0.002 5 mol·L~(-1)庚烷磺酸钠水溶液-乙腈(36∶37∶27),检测波长:250 nm,柱温:35℃,流速1.0 mL·min~(-1),进样量:10μL,以峰面积外标法定量。结果:甘草酸浓度在15.01~150.1μg·mL~(-1)范围内呈较好线性关系,回归方程为:Y=8.823X+35.60(R~2=0.999),平均回收率为100.6%,RSD=1.26%(n=6)。结论:此方法适用于测定复方甘草片中甘草酸的含量。
Objective: To improve the chromatographic conditions of China Pharmacopoeia and establish an HPLC method for the determination of glycyrrhizic acid in compound liquorice tablets. Methods:Shim-pack GIST C_(18)(4.6 mm ×250 mm,5 μm) column was used at a temperature of 35 ℃. The mobile phase was 0.025 mol·L~(-1)KH_2PO_4-0.002 5 mol·L~(-1)C_7H_(15)NaO_3S-acetonitrile(36∶37∶27).The flow rate was 1.0 mL·min~(-1) and the UV detection wavelength was at 250 nm.The sample injection volume was 10 μL.The external standard method of peak area was employed for the quantitative analysis. Results:A good linearity was obtained in the range of 15.01-150.1 μg·mL~(-1), the regressionequation was Y=8.823X+35.60(R~2=0.999), and the average recovery was 100.6%, with RSD of 1.26%(n=6). Conclusion:The method is good for the determination of glycyrrhizic acid in compound liquorice tablets.
