摘要
目的建立HPLC法测定石见穿中迷迭香酸和丹酚酸B的含量。方法采用ZORBAX SB-C18柱(5μm,4.6×250mm),以乙腈-0.1%甲酸溶液(20∶80)为流动相,检测波长为286nm。结果迷迭香酸在0.0154~1.286μg,丹酚酸B在0.0132~1.103μg范围内进样量与峰面积呈良好的线性关系(r=1.0000),平均回收率分别为102.7%和101.5%,RSD分别为0.8%和1.7%。结论本方法简便快捷、重复性好,为石见穿药材质量标准的建立提供研究基础。
OBJECTIVE To establish a method for determining the content of rosmatinic acid and salvianolic B in Herba Salvia chinensis.METHODS The HPLC method was adopted,the separation was performed on a column ZORBAX SB-C18(5μm,4.6×250mm),acetonitrile-0.1% formic acid(20∶80) was used as the mobile phase,and the determination wavelength was at 286 nm.RESULTS Both rosmatinic acid and salvianolic B showed a good linearity at the range of 0.0154~1.286μg and 0.0132~1.103μg(r=1.0000),respectively;the average recoveries of rosmatinic acid was 102.7% with RSD 0.8%(n=6),whereas 101.5% for salvianolic B with RSD 1.7%(n=6).CONCLUSION The method was simple,accurate,reliable and repeatable;it can provide a basis for quality evaluating for Herba Salvia chinensis as medicinal materials.
