摘要
目的:探讨归脾汤对化疗相关性疲劳模型TNF-α分泌的调节效应。方法:6周龄SPF级C57BL/6小鼠,雌雄各半,共80只,随机分为4组,每组20只,即空白对照组(A组)、化疗疲劳组(B组)、香菇多糖组(C组)和归脾汤组(D组)。实验前正常预养1周。将除A组以外的小鼠腹腔注射5-氟尿嘧啶(5-Fu),剂量为40mg/kg,每天给药1次,连续5天。同时,空白对照组给予生理盐水腹腔注射。造模前两天C组给予香菇多糖、D组给予归脾汤灌胃,A组和B组等量生理盐水灌胃,共14天。期间测量体重和抓力。第15天处死动物取腹腔巨噬细胞、脾和胸腺,测定TNF-α的分泌水平。结果:归脾汤可增加小鼠的体重、抓力以及胸腺指数和脾指数,降低TNF-α的分泌水平。结论:归脾汤可改善化疗引起的相关性疲劳,可能与降低TNF-α的分泌有关。
        Objective:To investigate the regulatory effect of Guipi Decoction on TNF-αsecretion in chemotherapy-induced fatigue model.Methods:Eighty six-week-old SPF C57 BL/6 mice were randomly divided into four groups,namely normal,group(group A),CIF group(group B),lentinan group(group C)and Guipi Decoction group(group D).The mice were injected 5-fluorouracil(40 mg/kg)i.p.daily for five days except group A,group A was given intraperitoneal injection of saline.At the same time,group C was given lentinan,group D was given Guipi Decoction,and group A and B were given saline for 14 days.Body weight and grasping power were measured during the experimental period.On the 15 th day,the mice were sacrificed to inspect the TNF-αlevels of peritoneal macrophages as well as spleen and thymus index were measured.Results:Guipi Decoction can increase the body weight of mice,grasping power and thymus index and spleen index,decrease the secretion level of TNF-α.Conclusion:Guipi Decoction can improve CIF caused by 5-Fu,may be associated with reduction of the TNF-α Secretion.
