化学计量学辅助高效液相色谱-二极管阵列检测器同时测定中药枳实中的六种黄酮类化合物

摘要
黄酮类化合物广泛存在于植物体内,具有多种生理活性,如抗炎、降血压、抗心律失常、抗氧化等。文献报道的对于黄酮类成分的定量分析大多基于液相色谱串联各种检测器,但这些方法往往存在耗时,预处理复杂等问题。本文采用高效液相色谱串联二极管阵列检测器(HPLC-DAD)与基于交替三线性分解算法(ATLD)的二阶校正方法结合的策略,同时测定来自不同产地枳实中的活性成分黄酮类化合物。通过简单的流动相,六种黄酮类物质在8分钟内全部洗脱出。凭借"二阶优势",该方法能解决色谱分离过程出现的峰重叠、未知干扰和基线漂移问题,使枳实中六种黄酮类物质的定量分析在相对较短的时间内完成。实验结果表明,该方法具有简单,快速的优点,在复杂体系多组分的同时定量分析中具有广阔的应用前景。
Flavonoids have been demonstrated to indicate a wide range of beneficial properties such as anti-inflammatory,antihypertensive,antiarrhythmic and antioxidant activity.Most quantitative methods in literatures are based on liquid chromatography with different detectors,but there are some drawbacks of time-consuming and complicated pretreatment.A novel strategy that combines high performance liquid chromatography with diode array detector(HPLC-DAD) with the second-order calibration method based on the alternating trilinear decomposition(ATLD)algorithm was proposed to quantify flavonoids in Aurantii Fructus Immaturus samples from different places.This method proved to be a useful tool for handling the overlapped peaks,uncalibrated interferences and baseline drifts existing in the process of chromatographic separation,which benefited from the "second-order advantages",making the determination of six flavonoids within 8 min under a simple mobile phase condition.Experimental results indicate the proposed method is fast and simple and has broad prospects for simultaneous determination of multi-components in complex systems.
