莫荷罗非鱼与其亲本间的DNA甲基化差异分析

摘要
运用甲基化敏感扩增多态技术分析橙色莫桑比克罗非鱼、荷那龙罗非鱼及其杂交子一代莫荷罗非鱼的皮肤、肌肉和鳃等11种组织的DNA甲基化水平差异,并简单探讨了杂种优势形成过程中甲基化模式的改变。采用16对引物进行选择性扩增,经聚丙烯凝胶电泳带型分析结果显示:莫荷罗非鱼及其亲本在不同组织的甲基化水平之间存在特异性,不同亲子代罗非鱼在相同组织的甲基化程度亦存在差异;橙色莫桑比克罗非鱼、荷那龙罗非鱼以及莫荷罗非鱼各组织平均总甲基化水平分别为38.03%、32.21%和29.77%,杂交种的甲基化水平低于双亲;莫荷罗非鱼基因组除了完全或部分继承双亲的甲基化模式之外,24.92%胞嘧啶位点发生去甲基化和超甲基化,且去甲基化位点多于超甲基化位点。结果表明杂交罗非鱼基因组DNA的低甲基化特征和甲基化模式的重调可能与杂种优势的形成有关,实验结果为进一步揭示罗非鱼的杂种优势形成机制奠定表观遗传学基础。
The methodof methylation sensitive amplified polymorphism(MSAP) was used to analyze the DNA methylation difference of eleven tissues of Oreochromismossambicus,Oreochromishornorumand "Mo-Ho" hybrid tilapia(O.mossambicus♀×O.hornorum♂),such as skin,muscle and gill etc, and to discuss the change of methylation pattern during the hybrid tilapia process of heterosis formation. In this study, 16 pairs of primers were used to amplify selectively.The results of electrophoretic banding analysis showed that there weredifferencesin the methylation levels among thesedifferent tissues andbetween thesame tissuesfrom "Mo-Ho"hybrid tilapia and itsparents; The average methylation level of these tissues from O.mossambicus,O.hornorumand "Mo-Ho"hybrid tilapia was 38.03%, 32.21% and 29.77%, respectively. The methylation level of hybrids was lower than the parents. Excepting for inheriting the methylation pattern from parents completely or partially, 24.92% cytosine demethylation and hypermethylation were found in the genome of hybrid tilapia, and the number of demethylation sites wasmore than that of hypermethylation sites.
