人工诱导鳗鲡(Anguilla japonica)成熟过程中血清及性腺内类固醇激素含量的变化

采用超高效液相色谱—串联质谱联用(UPLC-MS/MS)测定分析鳗鲡发育成熟过程中血清内六种类固醇激素:睾酮(T)、17α-羟基孕酮、17α,20β-二羟基-4-孕烯-3-酮(DHP)、雌酮(E_1)、雌二醇(E_2)、雌三醇(E_3)以及性腺中四种类固醇激素(T、DHP、E_2、E_3)激素的含量变化。结果表明,随着鳗鲡性腺发育,实验组鳗鲡血清及性腺中类固醇激素的含量呈逐步增加的趋势,其中,实验组鳗鲡血清中除E_3(难产时含量最高)外,其它5种类固醇激素含量均在产卵时最高;实验组鳗鲡性腺内E_2及DHP含量在难产时最高。对照组鳗鲡血清内E_1、E_3以及性腺内E_2、E_3在整个实验周期均未检测到,对照组激素含量均显著低于实验组。本方法具有灵敏、准确、稳定、回收率高的特点,结果揭示了鳗鲡性腺发育过程中性类固醇激素的变化,对鱼类性腺发育过程中体内激素变化的深入了解具有参考意义。
This study provides the method of ultra-performance liquid chromatography-tandem mass spectrometry for target steroid quantitation in the process of the Anguilla japonica maturation. We developed and validated the UPLC–MS/MS method to simultaneously determine6 steroids in serum samples:in gonad samples. The results showed that with the hormone injection,steroid hormones in serum and gonad tissue of experimental group Anguilla japonica were gradually increased. Among them, the content of 5 steroid hormones in serum is the highest when it spawned except for E_3(highest whendystocia). The content of E_2 and DHP in gonad tissue of experimental groups was the highest when dystocia.
