碱性成纤维细胞生长因子抑制过氧化氢诱导的骨髓间充质干细胞的凋亡

摘要
目的观察碱性成纤维细胞生长因子(basic fibroblast growth factor,b FGF)对过氧化氢(H2O2)诱导的骨髓间充质干细胞(Bone marrow mesenchymal stem cells,BMSCs)凋亡的保护作用,并探讨其机制。方法用b FGF干预H2O2处理的BMSCs,MTT法检测b FGF处理的BMSCs活性,LDH试剂盒检测细胞培养液乳酸脱氢酶(LDH)漏出量,TUNEL法检测细胞凋亡率,ELISA法检测细胞上清液中IL-6和TNF-α的表达及细胞中Bcl-2/Bax比值。结果 b FGF能显著改善细胞活性(P<0.05),降低LDH漏出量及凋亡率(P<0.05),减少细胞上清液中IL-6和TNF-α的表达(P<0.05),上调细胞的Bcl-2/Bax比值(P<0.05)。结论 b FGF可能通过减少细胞IL-6和TNF-α的表达以及上调Bcl-2/Bax比值,从而增强BMSCs的抗H2O2损伤能力。
Objective This study was designed to investigate protective effect and potential mechanisms of basic fibroblast growth factor(b FGF) against hydrogen peroxide(H2O2)-induced apoptosis of bone marrow mesenchymal stem cells(BMSCs).Methods b FGF treated BMSCs exposed to H2O2.MTT assay was used to detect the cell viability,LDH assay was performed to text lactate dehydrogenase leakage,DAPI staining and TUNEL assay was used to check apoptosis,and ELISA assay was served to detect the expression of interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) and Bcl-2/Bax ratio.Results b FGF treatment significantly improved viability of BMSCs(P<0.05),attenuated LDH leakage and apoptosis(P<0.05),decreased the levels of TNF-α and IL-6 and increased expession ratio of Bcl-2/Bax in H2O2-induced BMSCs(P<0.05).Conclusion b FGF increases the anti-apoptotic ability of BMSCs by decreasing the levels of TNF-α and IL-6 and increasing expession ratio of Bcl-2/Bax.
