几种鲍鱼性腺多糖提取方法的对比

摘要
本研究以雌性鲍鱼性腺为原料,比较了热水浸提、超声、微波、酶法、超声辅助酶及微波辅助酶六种提取方法对鲍鱼性腺多糖的影响。红外光谱及1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生化-HPLC分析表明,六种提取方法所得多糖均具备多糖的特征吸收峰且单糖组成相同,主要为葡萄糖醛酸、木糖、甘露糖、岩藻糖和葡萄糖。微波辅助木瓜蛋白酶法及酶法硫酸多糖提取率较高,分别为12.02±0.43%,12.01±0.15%,但前者总糖提取率较后者提高了48.94%;超声辅助木瓜蛋白酶法总糖提取率最高,为11.89±0.14%,较传统热水浸提法和酶法分别提高了118.83%,55.06%,但是其所得多糖的硫酸根含量最低,仅为4.93±0.02%。本研究综合评价了不同提取方法对鲍鱼性腺多糖的影响,发现超声辅助木瓜蛋白酶法有利于总糖的提取,而微波辅助木瓜蛋白酶法有利于硫酸多糖和总糖的提取。
In the study,effects of different extraction methods on the polysaccharides from female abalone gonad(AGP) were investigated.Six different extraction methods were used to extract AGP,which included hot water extraction(HWE),ultrasonic wave-assisted extraction(UAE),microwave-assisted extraction(MAE),enzyme-assisted extraction(EAE),ultrasonic-assisted enzyme extraction(UAEE) and microwave-assisted enzyme extraction(MAEE).FT-IR and l-Phenyl-3-methyl-5-pyrazolone(PMP) derivatization followed by HPLC analysis indicated that the AGP obtained by six extraction methods had the characteristic absorption peaks of polysaccharide,and the monosaccharide composition of them were similar,which mainly included GlcA,Xyl,Man,Fuc and Glc.The yield of sulfated polysaccharides by MAE and EAE were 12.02%±0.43%and12.01%±0.15%,respectively,which were higher than that by the other methods.While the yield of total sugar by MAE was increased by 48.94%compared to EAE.The yield of total sugar by UAE,11.89%±0.14%was the highest which was increased by 118.83%,55.06%compared to conventional HWE and EAE,respectively,while the AGP obtained by it contained the lowest content of sulfate radical which was only 4.93%±0.02%.In conclusion,the study evaluated comprehensively the effects of different extraction methods on AGP,and it was found that UAEE was useful to the extraction of total sugar,while MAEE was useful to the extraction of sulfated polysaccharides and total sugar.
