基于高通量测定肉鸡回肠微生物多样性及PICRUSt基因预测分析

摘要
本试验旨在研究健康肉鸡回肠的微生物群落结构及功能。采用高通量测序技术对15只鸡的回肠内容物进行测序,按照97%的相似度归类OTUs,去除序列数量少的OTUs(n<2),然后进行生物信息学分析及PICRUSt基因预测。结果表明:1)共获得424 062条tags,并归类于1 002个非单OTUs,样品平均tags为28 271±8 855条,平均OTUs为261±82个。2)所有样品共含有20个菌门,160个菌属。其中,共享核心菌门4个,分别是厚壁菌门(85.35%)、变形菌门(6.09%)、蓝细菌门(2.15%)和放线菌门(0.16%)。共享核心菌属19个,占样品总微生物总量的93.75%。3)鸡回肠预测得出328个功能。从前20功能热图可知,各样品的效能存在差异。由此可见,鸡回肠微生物群落复杂,但主要菌群相对稳定。本试验为进一步研究鸡回肠微生物群落结构与功能提供参考依据。
The aim of this study was to analyze the composition and function of healthy chickens ileum microbial community.The samples of chickens ileum microbial community were investigated by high-throughput sequencing.OTUs were picked by using de novo OTU picking protocol with a 97%similarity threshold and remove the sequences less than two in all samples.The results were analyzed by bioinformation software and PICRUSt functional profiles.Results showed that,1) a total of 424 062 tags were assigned to 1002 non-singleton OTUs from 15 samples.Each sample has28271±8855 tags and 261±82 OTUs on average.2) In total,20 phyla and 160 genera were detected.4 phyla were contained in all samples.They are Firmicutes(85.35%),Proteobacteria(6.09%),Cyanobacteria(2.15%) and Actinobacteria(0.16%).Nineteen genera were contained in all samples,which consisted 93.75%) of the overall bacteria community.3) 328 functions were detected in 15 samples.Heat-map of the first 20 functions showed that there are differences in efficiency of each sample.Therefore,our research revealed that the chicken ileum is complex for the phylogenetic diversity of the microbial communities,but the main microbial communities is relatively stable.The findings of this study will provide basic information and lay the foundation of the research in composition and function of the microbiota of chicken ileum.
