延胡索乙素对镇痛作用中5-羟色胺表达的影响

摘要
目的探讨延胡索乙素(tetrahydropalmatine,THP)对福尔马林足底致痛模型大鼠的镇痛作用以及在脊髓水平上的镇痛作用机制。方法以雄性SD大鼠为研究对象,分为正常组、模型组、阳性对照组(布洛芬组)和实验组(延胡索乙素组)等四组,在大鼠右后足足底部皮下注射4%的福尔马林100μL建立福尔马林足底致痛大鼠模型,并利用行为学指标、c-Fos蛋白(c-Fos Protein)表达检测方式来体现模型制备的准确成功率,随后又通过免疫组织化学法,蛋白质免疫印迹法等方法观察福尔马林足底致痛模型在不同的时间(30、60、90min)段,对L4-5腰段脊髓灰质后角内c-Fos蛋白、5-羟色胺的表达情况。结果福尔马林足底致痛模型大鼠的L4-L5脊髓灰质后角内c-Fos蛋白表达率60min时达到高峰,并确定福尔马林足底痛大鼠模型制备成功;延胡索乙素中剂量或高剂量组的c-Fos蛋白表达量、5-羟色胺表达量明显低于布洛芬组。结论延胡索乙素对由福尔马林引起的疼痛效应具有一定的抑制作用,且中、高剂量的延胡索乙素的抑制作用比布洛芬的效果更加显著,该作用可能是通过调节5-羟色胺来实现。
Objective To investigate the analgesic effects of tetrahydropalmatine(THP) on rat model with formalin foot pain and its analgesic mechanism on spinal cord.Methods Male Sprague Dawley(SD)rats were divided into four groups:normal group,model group,ibuprofen group and tetrahydropalmatine group.Rat models with formalin foot pain were established by subcutaneous injection in the right plantar hind paw with 4% formalin 100μl,and the success rate of rat model was judged by detecting the behavioral index and expression of c-Fos protein.The expression of c-Fos protein and 5-hydroxytryptamine(5-HT) in the posterior horn of L4-L5 lumbar spinal cord were detected by immunohistochemistry and Western blotting in different time points(30,60,90 min) on rat model with formalin foot pain.Results The expression rate of c-Fos protein in L4-L5 lumbar spinal cord posterior horn of rat model with formalin foot pain was reached peak at 60 min,which indicates that rat models with formalin foot pain were successfully prepared.The expression of c-Fos protein and 5-HT in the medium dose or high dose group of tetrahydropalmatine were significantly lower than those in the ibuprofen group.Conclusion THP had some inhibiting effects for formalin-induced pain,and the inhibitory effects of medium and high dose of THP were better than that of the ibuprofen,and its effects may be implemented by the 5-HT.
