病毒性脑炎和精神分裂症患者血液BDVp40基因片段的检测

摘要
目的探讨贵州省病毒性脑炎(Viralencephalitis, VE)、精神分裂症(Schizophrenia, SCH)患者博尔纳病病毒(Borna disease virus, BDV)的感染情况
     方法采用荧光定量巢式逆转录酶聚合酶链反应(FQ-nRT-PCR)检测来自贵州省遵义市、铜仁地区及毕节地区68例病毒性脑炎患者、38例精神分裂症患者、84名健康对照者外周血单个核细胞(peripheral blood mononuclear cells, PBMCs)中BDVp40基因片段。对阳性扩增产物进行基因序列测定,用软件对基因序列进行同源性分析
     结果68例病毒性脑炎患者外周血液单个核细胞中检出BDV p40基因片段阳性7例,遵义市6例、毕节地区1例。阳性率为10.29%(7/68)。38例精神分裂症患者外周血液单个核细胞中检出BDV p40基因片段阳性3例,阳性率为7.89%(3/38),遵义市2例、毕节地区1例。84名健康人外周血液单个核细胞中未检出BDV p40基因片段阳性。病毒性脑炎与健康人比较阳性率有显著性差异,P=0.009;精神分裂症与健康人比较阳性率有显著性差异,P=0.048。基因片段测序及同源性分析结果为:在病毒性脑炎和精神分裂症中检出的BDV p40基因片段阳性产物核苷酸序列一致,同源性100%。病毒性脑炎和精神分裂症患者阳性产物核苷酸序列与GeneBank提供的strain V/FR毒株比较同源性为95%,有5个位点出现一致性沉寂突变(nt276T-C, nt322 T-C, nt348 A-G, nt351 C-T, nt369 C-T);与H1766病毒株比较同源性为96%,有4个位点出现一致性沉寂突变(nt276T-C, nt322 T-C, nt348 A-G, nt351 C-T),与He80/FR病毒株比较同源性为98%,有2个位点出现一致性沉寂突变(nt322 T-C,nt348 A-G)。
     结论贵州省遵义市、毕节地区病毒性脑炎患者检测到BDV-P40基因片段,病毒性脑炎存在BDV感染,病毒性脑炎可能与BDV感染有关。贵州省遵义市、毕节地区精神分裂症患者检测到BDV-P40基因片段,精神分裂症存在BDV感染,精神分裂症可能和BDV感染有关。贵州省遵义市、毕节市的精神分裂症和病毒性脑炎患者BDV-P40基因序列相同,BDV感染可能来自相同的感染源。
Objective To investigate the Borna disease virus (BDV) infection of viral encephalitis (VE) and schizophrenia (SCH) in Guizhou Province.
     Methods FQ-nRT-PCR was applied to test P40 fragments of Borna disease virus in peripheral blood mononuclear cells (PBMCs) from 68 patients with VE,38 patients with SCH and 84 healthy subjects. Gene sequences of positive products were tested; the test results were analyzed by software.
     Results The PBMCs from 68 patients suffering from VE were tested in P40 fragments of BDV.7 cases were positive,including 6 in Zunyi City and 1 in Bijie region. The positive rate was 10.29%(7/68). The PBMCs from 38 patients suffering from SCH were tested in P40 fragments of BDV.3 cases were positive, including 2 in Zunyi City and 1 in Bijie region. The positive rate was 7.89%(3/38).84 healthy subjects were not detected in PBMCs for BDV p40 gene.There was a significant difference in positive rate between viralencephalitis patients and healthy subjects (P=0.009). There was a significant difference in positive rate between schizophrenia patients and healthy subjects(P=0.048). The results of gene sequence and homology analysis were that two kinds of VE and SCH patient's DNA sequence were the same. its homogeneity was 100%. Compared with the strain V, the homogeneity of nucleotides sequences of the patient was 95%, and 5 (nt276 T-C, nt322 T-C, nt348 A-G, nt351 C-T, nt369 C-T) nucleotides occurred silent mutations. Compared with the H1766, the homogeneity of nucleotides sequences of the patient was 96%, and 4 (nt276T-C, nt322 T-C, nt348 A-G, nt351 C-T) nucleotides occurred silent mutations. Compared with the He80/FR, the homogeneity of nucleotides sequences of the patient was 98%, and 2 (nt322 T-C, nt348 A-G) nucleotides occurred silent mutations.
     Conclusions BDV p40 gene fragments were detected in patients suffering from viral encephalitis in Zunyi City and Bijie region of Guizhou Province. Viral encephalitis were infected by BDV。VE may be interrelated with BDV infection. BDV p40 gene fragment were detected in patients with schizophrenia in Zunyi City and Bijie region of Guizhou Province. Viral encephalitis were infected by BDV。SCH may be interrelated with BDV infection. The BDV P40 gene fragments in patients with viral encephalitis and schizophrenia in Zunyi City and Bijie region of Guizhou Province were the same, which may come from the same source of infection.
