小刺猴头多糖对大鼠慢性萎缩性胃炎的实验研究

摘要
小刺猴头多糖(Hericium Caput medusae Polysaccharide,HP)是以小刺猴头子实体包括菌丝体为原料采用现代方法提取而成。小刺猴头别名针猴头菌,是猴头菌科真菌小刺猴头的子实体和发酵菌丝体。民间以子实体入药,有利肠胃、助消化的作用,可用于治疗慢性萎缩性胃炎、浅表性胃炎、消化性胃溃疡和十二指肠球部溃疡及其它原因引起的胃痛、胃酸过多等疾病。为探讨小刺猴头多糖对慢性萎缩性胃炎的治疗作用及其机制,本文进行了如下几方面的研究:
     1.采用2%水杨酸钠和30%酒精混合溶液灌胃,并结合劳累、饥饱失常等多因素方法复制大鼠慢性萎缩性胃炎模型,观察小刺猴头多糖对慢性萎缩性胃炎的影响。结果表明:小刺猴头多糖能使胃液中游离酸度和蛋白酶活性增强,血清胃泌素含量降低,血浆中cAMP含量升高而cGMP含量有所降低。病理检查结果表明,小刺猴头多糖能明显抑制胃窦及胃体粘膜萎缩变薄。
     2.采用碳末法进行小鼠小肠推进运动实验,观察小刺猴头多糖对小鼠胃肠推进功能的影响。结果表明小刺猴头多糖能明显增强小鼠胃肠的推进功能。
     3.采用耳肿胀法和扭体法观察小刺猴头多糖的抗炎镇痛作用。结果表明:小刺猴头多糖能明显的抑制二甲苯引起的急性炎症及冰醋酸导致的扭体反应,有良好的抗炎镇痛作用。
     实验结果表明:小刺猴头多糖能够改善慢性萎缩性胃炎的胃内环境;降低过高的血清胃泌素含量,升高血浆中的cAMP的含量,对胃粘膜有很好的保护作用;并且小刺猴头多糖能明显增强胃肠推进功能,有明显的抗炎镇痛作用。以上可能是小刺猴头多糖治疗慢性萎缩性胃炎,缓解其临床症状的理论依据。
Chronic atrophic gastritis (CAG) is a common disease of digestive system, which demonstrates pathological changes in focal and global gastric mucosa, with shrinkage and functional decrease in different level for gastric secrete. At present, CAG, especially with intestinal metaplasia (IM) and/or atypical hyperplasia, is considered to be precancerous lesions of gastric cancer (PLGC). Researchers have been focusing on CAG to pursuit for corresponding pathogenesis and pathological mechanism, which is not clearly defined up to today. CAG, a high occurrence and canceration disease, is considered as a kind of disorder resulted from multiple factors. No therapeutical strategy has been successfully developed with modern medical protocals. However, in traditional Chinese medicine fields, many exciting practices have been conducted in treating spleen and stomach disorders, which indicate a meaningful potential for CAG therapy.
     Hericium caput medusae is a kind of precious bacterium, which can eat and be used as medicine. In private, Hericium caput medusae’s fruiting body is always used as a drug to treat CAG, superficial gastritis (SG), gastric ulcer (GU), duodenal ulcer (DU) and other gastropathies. The main components of the extracts from fruiting body after fermentation are polysaccharide, monosaccharide, protein, amino acids and so on.
