益胃散对功能性消化不良小鼠胃排空和血清胃泌素影响的实验研究

摘要
目的研究益胃散对功能性消化不良(functional dyspepsia,FD)模型小鼠胃排空和血清胃泌素(gastrin,GAS)的影响,并探讨其作用机制。
     方法将100只昆明系雄性小鼠随机分为4组:空白对照组,模型组,吗丁啉组,益胃散组,每组各25只。对除空白对照组外的小鼠按照饥饱失常+左旋精氨酸(L-arginine,L-Arg)综合造模法进行造模。造模结束后,禁食不禁水24h,每组随机抽取各10只小鼠灌胃半固体糊0.5 g/只,15 min后脱颈处死小鼠,剖开腹腔取胃,计算胃内残留率,观察胃粘膜病理组织学改变。各组其余小鼠停止造模,记录体重等一般状态,给与相应干预因素。在实验第26d给药结束后,禁食不禁水24h,灌胃半固体糊0.5 g/只,15 min后,留取血清,放射免疫法测定GAS,处死小鼠剖开腹腔取胃,计算胃内残留率。
     结果①采用饥饱失常+L-Arg综合造模的FD模型小鼠胃内残留率增加,与空白对照组比较有统计学意义(P<0.05),模型组小鼠血清GAS水平低于空白对照组(P<0.05),各组小鼠胃粘膜均无器质性病理组织学改变。②经益胃散治疗后,小鼠胃内残留率降低,与模型组比较有统计学意义(P<0.05),与吗丁啉组比较无统计学差异(P>0.05);益胃散组小鼠血清GAS升高,与模型组比较有统计学意义(P<0.05),与吗丁啉组比较无统计学差异(P>0.05)。
     结论益胃散是干预FD模型小鼠的有效方剂;实验结果提示益胃散有促进FD模型小鼠胃排空和提高FD模型小鼠血清胃泌素水平的作用;促进胃排空、提高血清胃泌素水平可能为益胃散治疗功能性消化不良的作用机理之一。
Objective: To study the effects of Yiwei San on gastric emptying and gastrin(GAS) in serum of mice with functional dyspepsia(FD) and investigate its mechanism.
     Methods: 100 KM male mice were randomly allocated to blank control group, model group, domperidone treatment group and Yiwei San treatment group, 25 of one group. Except for the mice from the blank control group, all other mice were made model by irregular food intake plus L-arginine(L-Arg) for 15 days. When the models were finished successfully, nothing but water was offered to the mice for 24 hours, then 10 mice of each group were chosen randomly and each of the mice was irrigated with semi-solid material of 0.5 gram. After 15 minutes' standing, all the chosen animals were executed, cut the belly open, took out of the stomach, calculated the residue rate of material in stomach and observed the histological change of gastric mucosa with light microscope. The other mice of each group were given corresponding intervention measures while the general condition of the mice was recorded, such as the weight. When the treatment was finished, all the mice without feeding anything but water in 24 hours were irrigated with the semi-solid material of 0.5 gram. After 15 minutes' standing, all the experimental animals were executed. We got serum or stomach specimen, and calculated the residue rate of material in stomach. Gastrin in the blood was detected by the method of radioimmunoassay.
     Results: Compared with that in the blank control group, the residue rate of material in stomach in the model group was distinctly statistical difference (P<0.05).The general condition and the histological change of gastric mucosa showed: all the mice had not organic diseases. The concentration of gastrin in serum in the model group was lower than that in the blank control group (P<0.05). Gastrin increased in the Yiwei San treatment group, significantly different from that in the model group (P<0.05). There was no significant difference between the domperidone treatment group and the Yiwei San treatment group (P>0.05). The residue rate of material in stomach in the Yiwei San treatment group obviously decreased compared with that in the model group (P<0.05). There was also no significant difference between the Yiwei San treatment group and the domperidone treatment group (P>0.05).
     Conclusion: The results have prompted that Yiwei San can accelerate gastric emptying of functional dyspepsia mice model. The results suggest that Yiwei San improved gastrin level in serum of functional dyspepsia mice model. The mechanism of treatment FD might be that Yiwei San has the effects of promoting the release of gastrin in serum and gastric emptying.
