摘要
本论文系统研究了拳参(Polygonum bistorta)和黄帚橐吾(Ligularia virgaurea)两种植物的化学成分。
     拳参(Polygonum bistorta)系蓼科(Polygonaceae)蓼属(Polygonum)植物,为多年生草本植物,主要分布于华北、西北及山东、江苏、湖北等地。其性苦、涩、微寒,具有清热解毒、消肿止血之功效,用于赤痢、热泻、肺热咳嗽、痈肿、口舌生疮、吐血、痔疮出血、毒蛇咬伤等症。据文献报道,该属植物普遍含有黄酮及蒽醌类化合物,此外还含有芪、萜、有机酸等化合物。
     黄帚橐吾(Ligularia virgaurea)系菊科(Compositate)橐吾属(Ligularia)植物,属于千里光族(Senecioneae),为多年生灰绿色草本植物,主要分布于西藏东北部,云南西北部,四川,青海,甘肃等省区,生长于海拔2600-4700米的河滩、沼泽草甸、阴坡湿地及灌木丛中。一般根茎入药,具有清热解毒,抗炎杀菌,化痰止咳,活血止痛等功效。据文献报道,该属植物化学成分以倍半萜和吡咯里西啶生物碱为主。
     本论文对拳参(Polygonum bistorta)和黄帚橐吾(Ligularia virgaurea)两种植物的化学成分进行了系统研究,采用常规的硅胶柱层析(CC),葡聚糖(Sephadex LH-20)柱层析,制备薄层层析(PTLC)和重结晶等实验手段对其进行了分离纯化,利用现代波谱技术(IR EI-MS,HR-ESI-MS,~1H NMR,~(13)C NMR,DEPT,~1H-~1H COSY,HMBC等)对分离的化合物进行了结构鉴定,并对部分化合物进行了抗菌活性研究。从拳参中分离得到20个化合物,鉴定了其中的17个,其中化合物1.1-1.13首次从该属植物中分离得到;从黄帚橐吾中分离得到21个化合物,鉴定了其中的20个,其中2个为结构新颖的倍半萜二聚体(2.1&2.2)。此外,对其中部分倍半萜类化合物进行了抗菌活性研究,试验结果表明倍半萜二聚体(2.2-2.4)具有一定的抗菌活性,而其倍半萜单体(2.6-2.8)及墨西哥烷型倍半萜(2.15&2.16)基本不具有抗菌活性。
The chemical constituents of Polygonum bistorta and Ligularia virgaurea have been studied in this thesis.
     Polygonum bistorta is the species of family Polygonaceae tribe Polygonum Cass., a perennial herb widely distributed in northeast and northwestern regions, as well as Shandong, Jiangsu, Hubei Provinces. The rhizome of the plant was used as a folk remedy for the treatments of dysentery, tussis, carbuncle, hemorrhoids haematemesis and snake bite. Previous chemical investigations of this genus indicated that flavones and anthraquinones were its main components. In addition, it also contains terpenes and organic acids.
     Ligularia virgaurea is perennial herbages of family Compositate tribe Senecioneae subtribe, mainly distributed in northeast of Tibet, northwesten of Yunnan Province, Sichuan, Qinghai and Gansu Province. Its roots have long been used as folk medicine in China, possessed with efficacy of anti-inflammation, anti-bacterial, relieving coughs and phlegm, mobilization of the blood circulation and soothing pains. Previous studies on this genus indicated that it contained sesquiterpnes and pyrrolizidine alkaloids.
     The chemical constituents of these two plants have been studied. The compounds were isolated by means of silica gel and Sephadex LH-20 gel columm chromatography, preparative thin layer chromatography and recrystallization etc. The structures of these compounds were elucidated by spectroscopic methods (IR, EI-MS, HR-ESI-MS, ~1H NMR, ~(13)C NMR, DEPT, ~1H-~1H COSY, HMBC etc.). Seventeen compounds are isolated and elucidated from Polygonum bistorta. Thirteen (1.1-1.13) of them were first reported in Polygonum genus. And twenty compounds were isolated and elucidated from Ligularia virgaurea and two of them were novel dimeric sesquiterpenes (2.1 & 2.2). Moreover, some compounds were tested for their anti-bacterial activity against Escherichia coli, Bacillus subtilis and Staphlococcus aureus. The result indicated that dimeric sesquiterpenes (2.2-2.4) exihibited weak anti-bacterial activity, while the monomer (2.6-2.8) and the mexicanan-type (2.15 & 2.16) sesquiterpenes did not have any anti-bacterial activity.
