丹参与红花水溶性成分配伍抗大鼠脑缺血再灌注损伤的作用研究

摘要
目的:探讨丹参与红花水溶性成分配伍对大鼠脑缺血再灌注引起损伤的保护作用。方法:SD大鼠随机分为假手术组、模型组、丹参组、红花组、丹参红花配伍组、尼莫地平组,采用大脑中动脉局灶性栓塞(MCAO)模型,观察丹参与红花水溶性成分配伍后对脑缺血再灌注损伤大鼠的神经功能,脑梗死体积,脑含水量和脑指数,脑组织SOD活力、MDA含量,脑组织中IL-1β、IL-6和TNFα等炎症因子水平,以及尼氏小体数等的影响。结果:丹参水溶性成分、红花水溶性成分能更明显地减轻MCAO模型大鼠的神经功能缺损症状,促进神经功能缺损症状的恢复;显著地缩小脑组织梗死;降低脑含水量和脑指数,减轻脑水肿;显著提高脑组织中SOD活力,降低MDA含量,降低缺血侧脑组织IL-1β、IL-6和TNFα等水平;抑制尼氏小体数的减少。丹参水溶性成分与红花水溶性成分组合配伍后效果更佳,且优于单用,对各项指标均有明显改善和提高。结论:丹参水溶性成分与红花水溶性成分配伍对大鼠脑缺血再灌注引起的损伤具有明显的保护作用。
        Objective: To observe the protective effects of compatibility water-soluble components of Miltiorrhiza and Saffloweron cerebral ischemia and reperfusion injury. Methods: The SD ratswere randomly divided into groups: sham-operated group, model group, the group of Miltiorrhiza, the group of Safflower, the group of compatibility of Miltiorrhiza and Safflower, and Nimodipine group. Using the focal middle cerebral artery embolization(MCAO) model,we observed targets including neurological deficit score,infarct volume,brain index,brain water content,activity of SOD,content of MDA, levels of IL-1β,IL-6 and TNF-α of ischemic brain tissue, and quantity of nissl body. Results:Water-soluble components of Miltiorrhiza and Safflower could significantly reduce the symptoms of neurological deficits, promote the recovery symptoms of neurological deficits,narrow infarct volume of brain tissue obviously, reduce brain index and brain water,raise the activity of SOD,reduce the content of MDA,levels of IL-1β,IL-6 and TNF-α of ischemic brain tissue and inhibit reducing or disappearing of nissl body. The effects of the compatibility of water-soluble components of Miltiorrhiza and Safflower was better than single use separately.Conclusions:The compatibility of water-soluble components of Miltiorrhiza and Safflower has significant protective effects on ischemic brain injury.
