Gastrin-releasing Peptide Receptor:Gene Cloning,Sequence Analysis and Tissue Distribution in Pigs

Gastrin-releasing peptide(GRP) is a mammalian bombesin(BN)-like peptide,which plays a number of important physiological functions via its receptor(gastrin-releasing peptide receptor,GRPR) in most animals.However,little is known about the gene encoding GRPR and its functions in pigs.In this study,we first cloned and analyzed the pig GRPR gene.Then systematically investigated the expression levels of GRPR mRNA by relative real-time PCR(RT-PCR),and analyzed the distribution of GRPR protein via immunohistochemistry(IHC) in pig tissues.Results shown that pigs GRPR gene cloned 1,487 bp,including one open reading frame(ORF) of 1,155 bp and encoded 384 amino acids.Significantly,the sequences of the gene and amino acid of GRPR were highly homology and conservative in many species.The RT-PCR results showed that pigs GRPR mRNA was mostly detected in the central nervous system(CNS)and peripheral tissues.In the CNS,GRPR mRNA was found in the cerebellum,hypophysis,spinal cord and hypothalamus;in the peripheral tissues,GRPR mRNA was mainly expressed in the digestive tissues,the reproductive organs,the lymphatic organs,muscles,fat and so on.Moreover,the IHC results shown that GRPR immunoreactivity was widely distributed in pigs tissues and organs,such as the respiratory and circulatory system,digestive system,urogenital system,endocrine system and lymphatic organs.These systematically molecular and morphological data not only describe the anatomical locations of GRPR in pigs,but also provide the theoretic foundations for further research of the possible physiological functions in pigs.
