胃痞舒分散片的研制及治疗CAG癌前病变机理的初步研究

摘要
本文对治疗慢性萎缩性胃炎的中药复方制剂胃痞舒分散片的研制进行了较
    为系统的研究。采用正交实验设计优选了黄芪、半枝莲的水提取工艺,应用吸附
    澄清技术对其提取液进行精制处理并优选了吸附剂的种类和用量;采用超微粉碎
    技术将薏苡仁超微粉碎,并依据药效学指标,确定薏苡仁微粉的临床用量;采用
    均匀实验设计优选了莪术的超临界CO2流体萃取工艺,并用正交试验设计对挥
    发油的包合工艺进行了筛选,采用衍射法对包合物进行了鉴别;对分散片的辅料
    进行了选择;黄芪、半枝莲精制液经喷雾干燥后的浸膏粉,薏苡仁超微粉、莪术
    油环糊精包合物,混合均匀,加入50%乙醇适量湿法制粒,烘干、整粒。将所得
    颗粒、PVPP辅料等混合均匀、压片。采用薄层色谱法对制剂中半枝莲、莪术、
    薏苡仁进行鉴别;采用薄层扫描法对黄芪中黄芪甲苷含量进行测定,并进行了方
    法学的研究。考察了制剂初步稳定性,结果表明,制剂稳定性良好。通过对血清
    中SOD、MDA含量及胸腺、脾重量的测定,表明胃痞舒分散片具有抗氧化、调
    节免疫等作用。通过检测血管生成因子的表达,表明胃痞舒分散片具有抑制血管
    生成因子的作用。
Systematical research of weishu dispersibletablets, which had clinical practices for many years and accurate effects, has been performed in the study. In terms of document retrievals and initial experiments, the extraction technology of Radix Astragali and Herba scutellariae barbatae was determined by orthogonal test, then The extraction was refined using suitable clarifier. the equivalent dosage of Semen coicis after its micronization, compared with clinical dosage, was screened by pharmacologic experiments; the supercritical fluid extraction condition of Rhizoma curcumae was selected by uniform-design with table U7(76) and The conditions of process of extraction and inclusion were optimized by larger amount and efficient inclusion rate in inclusion complexes of volatile oil using orthogonal test, and controlling the quality of extract powders dried by spraying drier , completing the form of dispersible tablet which is a difficult procedure by selecting the disintegrants and diluents . The quality standards of dispersible tablet were established. The Herba scutellariae barbatae,Semen coicis,Rhizoma curcumae were identified by TLC. Radix . Astragali were quantitatively analyzed by TLC. Moreover, we have explored the beneficial effect of weishu dispersibletablets on the models of precancerous lesions of gastric cancel in Wistar rats by the animal experiments as follows: the change of SOD and LPO in plasma,the weight changes of immune organs and the expression of the the angiogenesis factors .The preliminary stability and pharmacological experiments show
    
    
    
    that the product has stable and controllable quality as well as definite effect on Precancerous lesions of chroic atrophic gastritis.
