大蒜素对人牙周膜成纤维细胞白细胞介素-6表达的影响

摘要
目的:研究大蒜素干预下正常及炎症牙周膜成纤维细胞中白细胞介素-6(interleukin-6,IL-6)的表达,初步探讨大蒜素治疗牙周炎及根尖周炎的作用机制。
     方法:1、组织块贴附法体外培养人牙周膜成纤维细胞(hPDLCs)原代并传代。2、接种第5代hPDLCs,酶联免疫吸附法(ELISA)测定无脂多糖(lipopolysaccharides,LPS)预处理及有LPS预处理后,用大蒜素干预下hPDLCs中IL-6的含量。
     结果:无LPS预处理情况下,大蒜素9mg/l浓度组与对照组相比IL-6的含量减少(P<0.05),较低浓度组与对照组之间IL-6含量无显著差异(P>0.05)。LPS预处理后大蒜素各浓度组与对照组相比IL-6含量均明显降低(P<0.05),但各组间IL-6的含量无显著差异(P>0.05)。
     结论:1.体外细胞实验中发现,9mg/l大蒜素作用于正常牙周膜成纤维细胞时可抑制牙周膜成纤维细胞分泌IL-6。2.LPS预处理后,各浓度组大蒜素均可明显抑制牙周膜成纤维细胞分泌IL-6。3.通过实验对比发现,大蒜素可明显抑制LPS预处理后的牙周膜成纤维细胞分泌过量的IL-6,,减轻IL-6对牙周组织的病理作用,这对牙周炎的治疗具有积极的意义。
Objective:To investigate the interleukin-6(IL-6) expression in normal and inflammatory periodontal tissue with allicin intervention,and preliminarily discuss the mechanism of allicin in treating periodontitis and periapical periodontitis.
     Methods:1.Human periodontal ligament fibroblast cells(hPDLCs) were cultured in vitro for primary and passaging generation using tissue explant attachment.2.The fifth generation of hPDLCs was inoculated, adding with allicin at different concentrations.Consequently,the IL-6 content in hPDLCs with or without LPS pretreatment was detected using enzyme-linked immunosorbent assay(ELISA).
     Results:In non-LPS group,IL-6 reduced in the group with allicin at concentration of 9 mg / L compared with the control group(P<0.05), no significant difference was shown in the lower concentration group (P>0.05).In LPS pretreatment group,allicin can obviously decreased IL-6 in each group compared with the control group,but the intergroup IL-6 showed no significant difference(P>0.05).
     Conclusion:1.During the in vitro experiment,9 mg / L allicin can inhibit normal hPDLCs to secret IL-6.2.With LPS pretreatment,allicin with concentration 3 mg / L,6 mg / L and 9 mg / L can all inhibit hPDLCs secreting IL-6.3.The experimental comparison shows that allicin can markedly inhibit the inflammatory periodontal tissue to secrete excessive IL-6.Allicin bears a positive effection in the treatment of periodontitis by alleviating IL-6.
