Characterization of a new acidic NAD+-dependent formate dehydrogenase from thermophilic fungus Chaetomium thermophilum

Chaetomium thermophilum formate dehydrogenase was overexpressed in E. coli.

The pH optimum was at pH 5, which is unusual for formate dehydrogenases.

The enzyme showed moderate thermostability and considerable storage stability at +4 °C.

Molecular properties of dimer–dimer interface were analyzed by modelling.

NAD+-mediated metal-independent hydride shift appears to be the reaction mechanism.
