Determinants of DNA binding specificity of paralogous HU and IHF is proposed. Three major clades belonging to HU, IHFα and IHFβ found from phylogenetic study. Conformational restriction due to salt-bridge formation increases their specificity. Alanine increases conformational space of side chains increasing their promiscuity. Determinants can be applied to understand specificity and promiscuity in proteins.
