Most of the reported effects of TLR4 signaling in tumors are pro-carcinogenic. Cytokines of TNF, IL-6, IL-1 families induce cancer mostly via chronic inflammation. Proinflammatory cytokines shape immunosuppressive tumor-promoting microenvironment. Existing blockers of proinflammatory cytokines are being evaluated in cancer therapy.
