Adeno-associated virus–mediated d2 silencing as a novel gene therapy for endodontic disease is proposed. A polymicrobial-induced periapical mouse model is used. d2 depletion simultaneously reduced bone resorption and enhanced bone formation. d2 depletion reduced inflammatory over-response and prevented bone erosion. This study revealed the triple functions of d2 silencing in the treatment of periapical disease.
