菊苣根中菊苣多糖含量测定研究

摘要
目的:建立合格的菊苣多糖含量测定方法。方法:采用苯酚-硫酸显色法在492.5 nm处测定菊苣总糖含量,DNS(3,5-二硝基水杨酸)显色法在475.5 nm处测定菊苣还原糖含量,计算得到菊苣多糖的含量。结果:菊苣总糖含量测定:D-果糖在15.3~35.7μg/mL范围内线性关系良好,回归方程为A=0.019,8C-0.089,3,r=0.999,7;菊苣还原糖含量测定:D-果糖在60~140μg/mL范围内线性关系良好,回归方程为A=0.006,1C-0.086,7,r=0.999,6。结论:本实验确定的菊苣总糖和菊苣还原糖的含量测定方法简便可行,重复性好,精密度、准确度高,可用于菊苣根中菊苣多糖的含量测定,以完善菊苣的质量标准。
        Objective: To establish the mensuration of chicory polysaccharides content. Method: The total sugar content of chicory was determined at 492.5 nm by phenol-sulfuric acid colorimetry, the content of reducing sugar in chicory was determined at 475.5 nm by DNS colorimetry. The content of chicory polysaccharide was calculated. Results: D-Fructose in total polysaccharide has a good linearity in the range of 15.3 ~35.7 μg/mL,and the regression equation was A=0.019,8 C-0.089,3, r=0.999,7. D-Fructose in reducing sugar has a good linearity in the range of 60~140 μg/mL, and the regression equation was A=0.006,1 C-0.086,7, r=0.999,6. Conclusion: The method of determination of total polysaccharide and reducing sugar determined in this experiment is simple and effective, precise, reproducible, accurate, and it can be used for chicory polysaccharide content determination in chicory root, to improve the quality standard of chicory.
