纤细裸藻对五种抗生素的胁迫响应

摘要
裸藻是一种重要的资源微藻,富含丰富的氨基酸、不饱和脂肪酸、维生素和裸藻糖等营养物质,并且裸藻没有细胞壁,其营养成分相对于其他藻类更容易被人体、水产动物吸收,因此开展裸藻集约化培养及养殖环境优化具有十分重要的意义。为了研究裸藻的无菌化养殖,实验生态条件下,研究了不同浓度的5种抗生素(遗传霉素、氯霉素、青霉素、土霉素和链霉素)胁迫对纤细裸藻(Euglena gracilis)细胞密度及叶绿素水平影响。结果表明:在遗传霉素、土霉素和链霉素胁迫作用下,裸藻细胞密度显著降低(P<0.05),低于25ug·mL~(-1)的遗传霉素即可对裸藻生长与叶绿素合成产生抑制,200ug·mL~(-1)的土霉素和25、50 ug·mL~(-1)的链霉素对裸藻叶绿素合成有一定的促进作用;氯霉素和青霉素对裸藻生长有一定的促进效应,200 ug·mL~(-1)的青霉素可以显著提高纤细裸藻的细胞密度和叶绿素水平(P<0.05)。实验结果说明纤细裸藻对遗传霉素、土霉素和链霉素的敏感性较强,而氯霉素和青霉素可以作为裸藻无菌系和基因工程选择标记的备选抗生素。
As an excellent resource microalgae, The Euglena riched in complex nutrients,such as amino acid, unsaturated fatty acids, vitamins and so on.The nutrient of Euglena was easier to absorb by humans and aquatic animal than other algae, because the Euglena lacked cell walls. Therefore, it is very important to develop the intensive cultivation pattern and optimize the breeding environment of Euglena. In order to studied the sterile culture, the Euglena gracilis were subjected to five kinds of antibiotics. The microalga growth and chlorophyll-levels were analyzed under laboratory conditions. Our results showed that: The Geneticin G418, Oxytetracycline and Streptomycin had significant effect on the cells density(P<0.05). The G418 showed a significant inhibitory effect on microalga growth and chlorophyll-levels when the concentration of was blow 25ug·mL~(-1). The Oxytetracycline(the concentration was 200ug·mL~(-1))and Streptomycin(the concentration was 25ug·mL~(-1) and 50ug·mL~(-1)) could enhance the chlorophyll-a levels. To some degree, Chloramphenicol and Penicillin could promote growth. Penicillin(the concentration was 200 ug·mL~(-1)) significantly enhance cells density and chlorophyll-levels of Euglena gracilis(P<0.05). The results of the study showed that the Euglena gracilis was more sensitive to G418, Oxytetracycline and Streptomycin. While the Cm and Penicillin could be the alternative antibiotic for the axenic culture of Euglena gracilis and selective marker of genetic engineering.
