摘要
本研究采用illumina Miseq高通量测序技术,对接种大豆根瘤菌与不接种大豆根瘤菌的大豆根际土壤进行总细菌DNA的提取,对细菌16srRNA高可变区V4~V5进行序列分析,对土壤中的细菌群落进行比较性分析。以不施肥的大豆根际土壤为参照,在施入不同根瘤菌处理后,分析大豆根际土壤的细菌区系在门、纲、目、科、属、种的分类群方面是否有差异改变,从分子生物学的角度来揭示黑龙江黑土土壤环境中外源根瘤菌对土壤微生物环境的影响。结果显示:与不接种根瘤菌的秋季土壤为参照,接种适宜的根瘤菌制剂能够丰富大豆根际土壤细菌群落,而不适宜的根瘤菌制剂不能对根际土壤环境有大的影响。由此说明,外源接种大豆根瘤菌引起根际土壤细菌的改变可能需要外源根瘤菌与大豆根系形成共生固氮关系,否则其对土壤群落的影响有限。
        Using illumina Miseq high-throughput sequencing technology,the total bacterial DNA was extracted from soybean rhizobium inoculated with soybean rhizobium and non-inoculated soybean rhizobium,and the bacterial16 srRNA hypervariable region V4 ~ V5 was sequenced in soil. Taking the soybean rhizosphere soil without fertilizer as a reference,after applying different rhizobium,this paper analyzed whether the bacterial flora of soybean rhizosphere soil had different changes in phylum,class,order,family,genus and species taxa,and revealed the effects of exogenous rhizobium on soil microbial environment in Heilongjiang black soil from the perspective of molecular biology. The results showed that the inoculation of suitable rhizobial preparations could enrich the bacterial community of soybean rhizosphere soil,and the unsuitable rhizobial preparation could not have a great impact on the rhizosphere soil environment.
